目的  探討水通道蛋白1(AQP-1)在炎癥性胸腔積液大鼠胸膜上的表達及其與胸水形成的關系。方法  56只健康Wistar大鼠隨機分為對照組(8只)和胸膜炎組(48只)。采用角叉菜膠胸腔內注射復制炎癥性胸腔積液大鼠模型。胸膜炎組根據注入角叉菜膠后處理時間再分為6,12,24,36,48和72 h組,每組8只。測定各組大鼠胸腔積液量,對胸腔積液進行細胞計數和分類,采用HE染色觀察臟、壁層胸膜的病理變化,采用免疫組化染色檢測AQP-1在大鼠胸膜上的分布,分別采用RT-PCR和Western blot方法檢測臟、壁層胸膜AQP-1的mRNA和蛋白表達。結果  胸膜炎各時相組胸水量分別為2.10±0.22、4.10±0.15、4.40±0.36、3.20±0.27、2.60±0.18和0.12±0.02 mL。AQP-1主要表達在臟、壁層胸膜的間皮細胞和毛細血管內皮細胞。胸膜炎各時相組臟、壁層胸膜AQP-1表達均增強。胸膜炎組AQP-1 mRNA在壁層胸膜的表達于6 h開始升高,24 h達高峰;在臟層胸膜的表達于12 h開始升高,24 h達高峰。AQP-1蛋白表達的變化趨勢與mRNA表達一致。12 h和24 h組壁層胸膜AQP-1表達與胸腔積液量正相關(r=0.857,r=0.846,P均 lt;0.01);24 h臟層胸膜AQP-1表達與胸腔積液量正相關(r=0.725,P lt;0.05)。結論  胸膜炎胸腔積液時胸膜AQP-1表達增強,AQP-1可能參與炎癥性胸腔積液的轉運過程。
